The Trans Pennine Trail runs across North West England, threading through the Peak District on its way. Roughly a third of the route is off road, so you move between sealed lanes and rougher surfaces across the ride. The terrain climbs through the hills before dropping back down, and the line shapes itself around that central high ground.
Five days at a moderate pace is a reasonable plan. Resupply depends on the towns along the way, and the off-road sections can be slower going than the overall distance suggests. The Peak District section is where the route bites hardest.
